Malaria causes 400,000 deaths annually, primarily among young children in Africa. Efforts to reduce the burden of malaria are hampered by the emergence of resistance in both parasites against available anti-malaria drugs and in mosquitoes against insecticides. High levels of lactic acid in the blood, or hyperlactatemia, are one of the strongest independent predictors of death in affected children and adults. However, the defined excessive lactate accumulates has been linked to these deaths. The aim of this research is to identify the physiological and molecular mechanisms leading to hyperlactatemia and to establish that hyperlactatemia is not merely a consequence of severe disease, but rather a cause of death. Understanding malarial hyperlactatemia could lead to targeted therapies for high-risk groups and inform our understanding of hyperlactatemia in fatal diseases such as sepsis and cancer.
