Biography

The Ralphe laboratory focuses on the study of inherited forms of cardiomyopathy, with a particular interest in hypertrophic cardiomyopathy. The research employs induced pluripotent stem cells (iPSCs) derived from patients with specific mutations in cardiac myosin binding protein C to generate cardiomyocytes (CMs). These cells are used to study the basic pathophysiology related to specific disease-causing mutations. The lab additionally utilizes CRISPR/Cas9 gene editing and adenoviral transduction approaches to introduce correct specific mutations into cardiomyocytes. Functional data generated from iPSC-derived CMs grown in multicellular living human engineered cardiac constructs exhibit the basic contractile characteristics of intact human myocardium.
