The O’Donoghue lab investigates proteome diversity, generated protein modification mistranslation, and impacts on cancer and neurodegeneration pathways. Proteins involved in major human diseases undergo significant post-translational modification, with these modifications relaying chemical signals that alter cell fate and gene expression. The lab has established a leadership role in the area of genetic code expansion to investigate the role of disease-linked protein modifications. They engineer new methods to produce proteins with programmed modifications to elucidate the signaling pathways linked to disease. Recent breakthroughs have enabled programmed protein modification in human cells, including the development of novel fluorescent reporters to visualize errors in protein synthesis and mistranslation in live cells. The lab is currently leading efforts to reveal the ability of naturally occurring transfer RNA (tRNA) mutants to induce proteome-wide mistranslation models associated with neurodegenerative disease.
