Dr. Ted Lakowski is a professor and the director of the pharmaceutical analysis laboratory at the College of Pharmacy, Rady Faculty of Health Sciences, University of Manitoba. He joined the College of Pharmacy in 2012. His research focuses on drug discovery that targets epigenetic processes, examining how drugs influence gene expression mechanisms using LC-MS/MS techniques. His laboratory investigates the effects of disease states and novel drugs on patterns of epigenetic modifications in cultured cells, in vitro, and animal models through metabolomic approaches. Dr. Lakowski's lab has developed a validated LC-MS/MS method to quantify multiple histone modifications in cultured cells treated with histone deacetylase (HDAC) inhibitors, demonstrating that these inhibitors alter gene expression by decreasing levels of histone lysine demethylases. His team has also discovered a new class of anti-cancer drugs, DOT1L inhibitors, which affect the genome and cause changes in histone modifications and the expression of histone-modifying enzymes. Additionally, they identified acetylated lysine residues on myelin basic protein that may be involved in the pathogenesis of multiple sclerosis. Dr. Lakowski's lab is currently working on methods to measure multiple DNA modifications and is involved in projects measuring metabolites related to glioblastoma progression.
